摘要: 采用荧光光谱法和紫外-可见吸收光谱法研究了在模拟人体生理条件下不同温度时,血卟啉与人血清白蛋白结合反应的荧光猝灭光谱行为,根据Stern-Volmer方程和Lineweaver-Burk双倒数函数曲线计算了其结合常数在高温时要比低温时小;根据Fster能量传递原理计算出结合位点r=4.36,并根据热力学参数确定了它们之间的作用力类型主要为氢键和Van der Waals力,HP进入了HSA的疏水腔。

Abstract:The quenching reaction of hemoporphyrin with human serum albumin(HSA) was studied by using fluorescence spectra and absorption spectra.The formation constants of them were analyzed at different temperature according to Stern-Volmer equation and double-reciprocal equation, which are smaller at high temperature than at low temperature.The binding site was calculated (r=4.36) by Fster energy transfer mechanism, and the thermodynamic parameters were obtained.H-bond and Van der Waals are two main reactions between HP and HSA.
