Determination of 8-Hydroxy-2’-Deoxyguanosine in Urine with Protamine Using Resonance Energy Transfer Fluorescence Quenching by Sensitizing Effect of Triton X-100

Abstract:The optimum conditions for the resonance energy transfer between 8-hydroxy-2’-deoxyguanosine(8-OHdG) and protamine were studied,and a novel method for the determination of 8-OHdG has been developed based on the resonance energy transfer fluorescence quenching effect of protamine. The fluorescence quenching intensity of Ps at 301 nm was proportional to the concentration of 8-OHdG. The linear range of the calibration curve was 0.50-7.1 μg·mL-1 in the pH 8.2 Tris-HCl buffer solution with the detection limits of 0.18 μg·mL-1. The relative standard deviation was 6.2% and the average recovery was 104%(n=6). The proposed method is simple and sensitive,and has succeeded in analyzing the spiked urine sample with satisfactory results.
